Molecular Diagnosis Compared to Conventional Diagnosis for Rapid Detection of Resistant Strains of Candida Spp.

Yeast Candida spp. can cause serious infections, especially in patients of the hospital environment. Variation virulence profile clinical isolates and high mortality rates indicate importance rapid accurate identification, as well susceptibility testing for successful candidiasis treatment. A total 20 strains were isolated from Tertiary Hospital Northern Zone Ceará. PCR amplification followed by agarose gel electrophoresis (PCR-AGE) manual method (culturing on CHROMagar-Candida) VITEK automated used to test a fungal clinics sources. Five C. albicans, 2 tropicalis 4 parapsilosis which cultured determination MIC (Minimum Inhibitory Concentration). The best sensitivity rate was demonstrated automatic identification PCR-AGE, where all with 100% (20/20) completely identified. According data obtained, resistance amphotericin B (CIM ≥ 1 µg/ml) observed some parapsilosis. However, other antifungals, these same species presented (MIC ≤ 0.125 μg / ml; 8 ml). Isolates albicans sensitive antifungal agents sensitivity, although 75% showed tendency less B. In this study, we confirmed efficacy applicability PCR-AGE differentiation yeast samples.
